Potential Roles of Muscle-Derived Extracellular Vesicles in Remodeling Cellular Microenvironment: Proposed Implications of the Exercise-Induced Myokine, Irisin

چکیده

Extracellular vesicles (EVs) have emerged as key players of intercellular communication and mediate crosstalk between tissues. Metastatic tumors release tumorigenic EVs, capable pre-conditioning distal sites for organotropic metastasis. Growing evidence identifies muscle cell-derived EVs myokines potent mediators cellular differentiation, proliferation, metabolism. Muscle-derived cargo other biological modulators like microRNAs, cytokines, chemokines, prostaglandins hence, are likely to modulate the remodeling niches in vital sites, such liver adipose Despite scarcity support a direct relationship muscle-EVs cancer metastasis, their indirect attribution regulation niche establishment pre-metastatic homing can be put forward. This hypothesis is supported by role muscle-derived findings gathered from pathologies inflammation metabolic disorders. In this review, we present discuss studies that evidently potential roles events metastatic tumor microenvironment. We highlight contributions integrin-mediated interactions with an emerging myokine, irisin, EV-driven microenvironment Further research into progression imperative may hold promising advance our knowledge pathophysiology, therapeutic management cancers.

Extracellular Vesicles: Potential Roles in Regenerative Medicine

Extracellular vesicles (EV) consist of exosomes, which are released upon fusion of the multivesicular body with the cell membrane, and microvesicles, which are released directly from the cell membrane. EV can mediate cell-cell communication and are involved in many processes, including immune signaling, angiogenesis, stress response, senescence, proliferation, and cell differentiation. The vast...

Irisin, a novel myokine: potential role in obesity and diabetes

Obesity is a worldwide health problem that results in a signifi cant increased risk of morbidity and mortality. While we know that obesity is the consequence of a chronic imbalance between energy intake and energy expenditure, how energy expenditure is regulated in humans is not clearly understood. Recent publications suggest that specifi c depots of white adipose tissue can be converted to the...

Irisin – a myth rather than an exercise-inducible myokine

The myokine irisin is supposed to be cleaved from a transmembrane precursor, FNDC5 (fibronectin type III domain containing 5), and to mediate beneficial effects of exercise on human metabolism. However, evidence for irisin circulating in blood is largely based on commercial ELISA kits which are based on polyclonal antibodies (pAbs) not previously tested for cross-reacting serum proteins. We hav...

Emerging roles of extracellular vesicles in cellular senescence and aging

Cellular senescence is a cellular program that prevents the proliferation of cells at risk of neoplastic transformation. On the other hand, age-related accumulation of senescent cells promotes aging at least partially due to the senescence-associated secretory phenotype, whereby cells secrete high levels of inflammatory cytokines, chemokines, and matrix metalloproteinases. Emerging evidence, ho...
